In vivo measurement of the myelin g-ratio with histological validation

چکیده
PURPOSE: The myelin g-ratio, defined as the ratio between the inner and the outer diameter of the myelin sheath, is a fundamental property of white matter that can be computed from a simple formula (Fig. 1) relating the myelin volume fraction (MVF) to the fiber volume fraction (FVF) or the axon volume fraction (AVF). Recent studies have suggested that the sexual dimorphism in white matter development is due to a higher g-ratio (relatively thinner myelin) in adolescent boys. Additionally, in vivo imaging of the myelin thickness in multiple sclerosis could provide a real-time tool for tracking myelination in lesions, facilitating the development and evaluation of new therapeutic agents that promote remyelination. In this abstract, a unique combination of magnetization transfer, diffusion imaging and histology is presented, providing a novel method for validating the in vivo measurements of the myelin g-ratio.

Recent years have seen a growing interest in relating MRI measurements to the structural-biophysical properties of white matter fibers. The fiber g-ratio, defined as the ratio between the inner and outer radii of the axon myelin sheath, is an important structural property of white matter, affecting signal conduction. Recently proposed modeling methods that use a combination of quantitative-MRI ...

Introduction: Prior qualitative and quantitative studies have reported morphological changes that occur in nerve fibers and non-neuronal cells of peripheral nerve during the lifetime of the rats. Previous studies suggest that Aloe Vera has some beneficial and therapeutic effects such as anti-oxidant effects. In the current study, we have evaluated histological and histomorphometric changes of Sc...
متن کاملQuantitative Histological Validation of Diffusion MRI Fiber Orientation Distributions in the Rat Brain
Diffusion MRI (dMRI) is widely used to measure microstructural features of brain white matter, but commonly used dMRI measures have limited capacity to resolve the orientation structure of complex fiber architectures. While several promising new approaches have been proposed, direct quantitative validation of these methods against relevant histological architectures remains missing. In this stu...
متن کاملMapping an index of the myelin g-ratio in infants using magnetic resonance imaging
Optimal myelination of neuronal axons is essential for effective brain and cognitive function. The ratio of the axon diameter to the outer fiber diameter, known as the g-ratio, is a reliable measure to assess axonal myelination and is an important index reflecting the efficiency and maximal conduction velocity of white matter pathways. Although advanced neuroimaging techniques including multico...

Introduction: Demyelination is one of the main causes of neurological disability. It is the end product of numerous pathological processes, multiple sclerosis (MS) being the most common cause. More than 70% of the MS patients suffer from optic disturbances. This disease commonly affects the optic pathway, particularly the optic nerves and chiasm. Several attempts have been made to produce a ...
